Genome Explorations has expanded it portfolio of services by introducing genotyping using the Affymetrix GeneChip single nucleotide polymorphism (SNP) array sets. Our scientists have developed new SOP’s for genotyping services with total dedication to generating reproducible data sets of the highest quality. As with our expression protocols, our genotyping protocols are engineered with reproducibility and quality in mind and have helped us generated data sets that are published in premier tier one journals.

Gene expression and genotyping microarrays have revolutionized the drug discovery process by allowing for genome wide screening for novel therapeutic targets. Genome Explorations will go one step further and provide reliable data in a GLP compliant format for use in FDA approval studies.
Process
Starting with Between 0.5 and 1mg of good quality genomic DNA.
DNA Quality Check and Quantitation
Each DNA sample is quantitated using a dual beam spectrophotometer and a negligible amount is used to perform QC check using an Agilent Bioanalyzer 2100 "Lab on a Chip" system. Investigators will be notified immediately if the DNA is degraded or otherwise unsuitable for GeneChip application.
DNA Processing
DNA samples that pass QC are first digested with a restriction enzyme. After adaptor ligation a PCR reaction is set up to amplify 250 – 2000bp fragments. The resulting amplified DNA is then fragmented, denatured, and tagged using a terminal deoxynucleotidyl transferase end labeling reaction.
GeneChip Microarray Hybridization and Data Analysis
Labeled fragments will be hybridized to the requested array. Microarray data will then be analyzed according to the client’s specifications.
